What editor you set as a default for front-end user ? Are you sure that this editor has set proper permission access ? For test please also switch template to default one - we will know whether the problem comes from our product or from K2 extension.

If I change to the default ATOMIC template, I am able to save changes to a user description without getting the 404 error..

what was the result? I have the same problem with gk_magazine, K2 v2.6.6 and Joomla 3.

I don't get any details. Did you try to test this issue on default Joomla! template ? Maybe this is just problem with jQuery library, please try change jQuery source in k2 configuration from remote copy to local copy.
